The aim of the present study was to examine the contribution of the periodic pancreatic secretion (PPS) associated with a duodenal three-phased migrating myoelectric complex (MMC) to the intra-animal variability in protein electrophoretical profiles of pancreatic juice. The profiles of pancreatic juice proteins in the PPS trough and the PPS peak in the preprandial period, during milk feeding, and following intravenous and intraduodenal administration of CCK-8 were studied in six 2-week-old calves surgically fitted with pancreatic duct catheters, duodenal cannulas and 2 duodenal bipolar electrodes. Duodenal MMC recording served us with precise identification of PPS cycles. Juice protein samples, adjusted to 300 ng/ml, were applied to an automated SDS-polyacrylamide gel (12.5%) electrophoresis system. Gels were silver-stained, scanned and analysed for trace density (TD; band optical density x band height) by the computer. The separations varied in the number of bands, according to the phase of the PPS. In the PPS trough 17 protein bands were detected, and at the PPS peak, 21 bands. Moreover, within the detectable bands there were significant differences in the TD between the PPS trough and peak. Following feeding and CCK-8 infusion different numbers of protein bands were observed. In conclusion, by collecting pancreatic juice strictly in phase with its natural periodic secretion and analysing the juice proteins using a computerised approach, we have shown that pancreatic secretion is apparently not uniform. The differences concern the interdigestive as well as the prandial and CCK-8-stimulated composition of pancreatic juice.
